Desarrollo del cultivo y evaluación de la diversidad genética en la almeja rubia Venerupis rhomboides

Aquaculture development and genetic diversity assessment in the banded carpet-shell clam Venerupis r

Este proyecto coordinado aborda el desarrollo de las técnicas de cultivo en criadero y el análisis de la diversidad genética en la almeja rubia Venerupis rhomboides  Esta almeja constituye un importante recurso para el sector marisquero de Galicia al ser la cuarta especie de almeja en importancia en cuanto a capturas e ingresos. El desarrollo de su cultivo, preservando la diversidad genética de las poblaciones naturales, supondría un gran beneficio para el sector al posibilitar el aumento de producción. En la actualidad las vedas y el cupo de capturas es diferente según la zona donde se vaya a extraer obviando cualquier aspecto biológico de la especie. El conocimiento de sus ciclos de reproducción y larvario es necesario para una mejor gestión del recurso. También existen zonas improductivas que se podrían recuperar haciendo siembras con semilla procedente de criaderos.

 

Para desarrollar el cultivo de V. rhomboides en criadero se pondrán a punto las técnicas necesarias. Se comenzará con el estudio del ciclo gametogénico en el banco natural de donde se recogerán los reproductores abordando su histología y composición bioquímica. Se harán muestreos mensuales en un banco y otros puntuales en otros bancos de otra zona para comparar y poder marcar el ciclo anual de esta especie en Galicia. De estos mismos individuos se estudiará la patología y microbiología ya que según los trabajos realizados en el criadero para otras especies parece tener incidencia directa en los resultados de los cultivos. En relación al cultivo, se estudiarán: las condiciones de estabulación de los reproductores (temperatura, alimentación, flujos de agua), la inducción de la puesta tomando de referencia métodos efectivos en otros moluscos, el desarrollo embrionario (duración, descripción y composición del huevo) y  el cultivo larvario partiendo de larva D hasta postlarva estudiando los diferentes estadios larvarios (duración, crecimiento y supervivencias).

 

Para el análisis de la diversidad genética se contempla el desarrollo de marcadores moleculares basados en el ADN por no haber sido identificados previamente en V. rhomboides. Se propone la identificación de marcadores nucleares tipo microsatélite de uso habitual en los análisis genéticos relacionados con la gestión de recursos, cultivo y mejora genética de especies de interés en acuicultura por mostrar un elevado polimorfismo. También se propone examinar secuencias de ADN mitocondrial por resultar este marcador más sensible a los cambios de tamaño poblacional. El análisis exhaustivo de al menos 10 loci microsatélite y dos genes mitocondriales en muestras de al menos 100 individuos recogidas a lo largo de la costa gallega permitirá obtener datos robustos sobre la diversidad genética y diferenciación poblacional, necesarios para la gestión sostenible de este recurso, desarrollo del cultivo y para servir de referencia en el futuro en el monitoreo de la alteración de la diversidad genética debida a cambios ambientales naturales y antropogénicos y/o prácticas de extracción, cultivo y repoblación. Además, se pretende proporcionar asesoramiento genético al desarrollo del cultivo de V. rhomboides mediante el análisis de la diversidad genética de los reproductores utilizados en el criadero y de la semilla obtenida, así como su grado de diferenciación genética respecto a la población natural que proporcionó los reproductores. Mediante la puesta a punto de pruebas de paternidad se tratará de dilucidar la contribución de cada reproductor a la descendencia y detectar así posibles desequilibrios entre el tamaño real del censo y el tamaño efectivo de población que conduzcan a la pérdida de diversidad genética. Para facilitar el proceso de genotipado de microsatélites se optimizarán PCRs multiplex lo cual permitirá reducir coste y tiempo no sólo en el desarrollo del presente proyecto sino también en futuros análisis que se puedan llevar a cabo en el ámbito académico e industrial.

 

En términos generales, este proyecto pretende contribuir a la producción sostenible de V. rhomboides para que las  actividades del sector marisquero y de acuicultura, con gran repercusión socioeconómica e Galicia, no se vean comprometidas en el futuro.

This coordinated project aims the development of aquaculture techniques and the assessment of genetic diversity in the banded carpet-shell clam Venerupis rhomboides. This clam is an important resource for shellfish harvesters within Galicia because this species in the forth clam species in importance not only in income but also in captures. Aquaculture development by means of preserving genetic diversity of wild stocks would mean a great benefit due to the rise of its production. Currently close seasons and harvesting quotas are different between areas without bearing biological aspects in mind. Moreover, unproductive areas could be replenished with hatchery produced seed.

 

In order to develop the hatchery production of V. rhomboides different techniques will be tested. Firstly, a study of gametogenic cycle in a natural bed, where broodstock will be collected, will be carried out by means of studying histology and biochemical composition. Samples will be collected in a monthly basis from a natural bed and other occasional samples will be harvested from other wild stocks in order to compare and establish the annual gemetogenic cycle of this species in Galicia. These individuals will be used to undertake pathological and microbiological studies due to the relationship between them and the cultivation results based on previous results in hatcheries obtained with other bivalve species. Regarding to cultivation, there will be studied: broodstock holding conditions (temperature, feeding and water flow), spawning induction using effective methods developed for other mollusks, embryonic development (length, description and biochemical composition of the eggs) and larval culture from D-shaped veliger to postlarvae (length, growth and survival).

 

With regard to genetic diversity, molecular markers based on DNA will be developed because it has not been previously identified in V. rhomboides. We propose the identification of nuclear markers of the type microsatellite, which are currently used in genetic studies related to stock assessment, cultivation and genetic improvement in species of interest for aquaculture due to their high degree of polimorfism. We also propose the examination of mitochondrial DNA sequences due to the sensitivity of this marker to changes in population size. The exhaustive analysis of at least 10 loci and two mitochondrial genes in samples of at least 100 individuals along the Galician coast will allow obtaining solid data dealing with genetic diversity and population differentiation, which are needed for sustainable management of this resource, culture development. Also, it could serve as reference for the future monitoring of the genetic diversity changes caused by both anthropogenic and natural environmental changes and/or harvesting, cultivation and restocking. Moreover, we would like to provide genetic advice for the development of V. rhomboids culture by means of not only the analysis of the genetic diversity of the broodstock used at hatchery facilities and the seed produced in hatcheries facilities but also the degree of genetic differentiation in relation to the wild population where the broodstock were collected. By means of the development of paternity tests we will try to ascertain the contribution of each broodstock to descendants and by means of that try to detect imbalances between the real size of the census effective size of the population which lead to a loss of genetic diversity. In order to facilitate microsatellite sequencing PCR techniques will be optimized, which will allow to reduce costs and time not only for the development of this current project but also for analysis in the future in the academic and industrial fields.

 

In conclusion, this project aims to develop a sustainable production of V. rhomboids in order that the activities of the shellfish harvested and aquaculture, which are of great socioeconomic importance in Galicia, will not be put at risk in the future.
